Detects rat CXCL3/CINC-2 alpha / beta in direct ELISAs and Western blots. In direct ELISAs, greater than 50% cross-reactivity with recombinant rat (rr) CINC-1, rrCINC-2 beta, and rrCINC-3 is observed, less than 25% cross-reactivity with recombinant mouse (rm) KC, rmMIP-2, recombinant human (rh) GRO alpha, rhGRO beta, and rhGRO gamma is observed, and less than 10% cross-reactivity with rhIL-8 is observed.

Neutralization
Measured by its ability to neutralize CXCL3/CINC-2 alpha -induced chemotaxis in the BaF3 mouse pro‑B cell line transfected with human CXCR2. The Neutralization Dose (ND50) is typically 0.5-2.0 µg/mL in the presence of 0.05 µg/mL Recombinant Rat CXCL3/CINC-2 alpha.

Chemotaxis Induced by CXCL3/CINC‑2 alpha and Neutral-ization by Rat CXCL3/ CINC‑2 alpha / beta Antibody. Recombinant Rat CXCL3/ CINC-2a (Catalog # 516-CA) chemoattracts the BaF3 mouse pro-B cell line transfected with human CXCR2 in a dose-dependent manner (orange line). The amount of cells that migrated through to the lower chemotaxis chamber was measured by Resazurin (Catalog # AR002). Chemotaxis elicited by Recombinant Rat CXCL3/ CINC-2a (0.05 µg/mL) is neutralized (green line) by increasing concentrations of Goat Anti-Rat CXCL3/ CINC-2a/ beta Antigen Affinity-purified Polyclonal Antibody (Catalog # AF516). The ND50 is typically 0.5-2.0 µg/mL.

Background: CXCL3/GRO gamma/CINC-2/DCIP-1

CINC-2 alpha and -2 beta are mRNA splice variant isoforms that differ by only 3 amino acids at their amino-termini. These proteins are CXC chemokines most closely related to human GROs and mouse MIP-2. They are produced by granulation tissue, activated macrophages and fibroblasts. CINC-2 biological activities are mediated by CXCR1 or CXCR2.
