Activity
Recombinant Human AMSH is a Ubiquitin-specific deconjugating enzyme. Reaction conditions will need to be optimized for each specific application. We recommend an initial Recombinant Human AMSH concentration of 100-500 nM.  AMSH activity is partially or fully inhibited by DTT at concentrations above 1 mM.

Carrier Free

What does CF mean?

CF stands for Carrier Free (CF). We typically add Bovine Serum Albumin (BSA) as a carrier protein to our recombinant proteins. Adding a carrier protein enhances protein stability, increases shelf-life, and allows the recombinant protein to be stored at a more dilute concentration. The carrier free version does not contain BSA.

What formulation is right for me?

In general, we advise purchasing the recombinant protein with BSA for use in cell or tissue culture, or as an ELISA standard. In contrast, the carrier free protein is recommended for applications, in which the presence of BSA could interfere.

Background: AMSH/STAMBP

AMSH (STAM binding protein) is a zinc metalloprotease belonging to the JAMM (JAB1/MPN/Mov34) family of deubiquitinating enzymes (DUBs). This enzyme functions at the endosome, where it is involved in the sorting of various cell-surface receptors to lysozomes. AMSH cleaves K63-linked but not K48-linked polyubiquitin chains, and the in vitro activity of AMSH is greatly increased in the presence of STAM protein. This recombinant protein is full-length.
