    I had loaded 1μg of recombinant myostatin/GDF8 into a 4-15% gel, performed electrophoresis with MES buffer and blotted the gel onto 0.2μm nitrocellulose membrane. For the immuno-detection, I had used the R&D systems goat polyclonal GDF8 antibody (BAF788) at 0.1 or 0.25 µg/mL concentrations, and the R&D system anti-goat secondary antibody (HAF017) at 1:4000 dilution.

    Dot blot test: The protein samples were neat and not diluted. The samples were compared against three control dilutions from human liver cells (neat, 1:2, and 1:5) which were also targeted for GAPDH.
