Blockade of Receptor-ligand Interaction

In a functional flow cytometry test, biotinylated recombinant human IL-2 (100 ng/mL, Catalog # BT202) binds to HEK293 human embryonic kidney cell line transfected with human CD25/IL-2Ra. Binding is blocked by 2.5 μg/mL of Mouse Anti-Human CD25/IL-2Ra Monoclonal Antibody (Catalog # MAB10203).

Human PBMCs were treated with Recombinant Human IL-2 (202-IL,20 ng/mL) and TGF-beta (100-B,10 ng/mL) for 24 hours, then stained with (A) Rabbit anti-Human CD25/IL-2 Ra Monoclonal Antibody (Catalog # MAB10203) or (B) Rabbit IgG control antibody (MAB1050), followed by PE-conjugated Anti-Rabbit IgG Secondary Antibody (F0110) and Mouse anti-Human CD3 APC-conjugated Monoclonal Antiobdy (FAB100A). IL-2 binding to Human CD25/IL-2R alpha-transfected HEK293 Human Cell Line is Blocked by Human CD25/IL-2R alpha Antibody. In a functional flow cytometry test, Recombinant Human IL-2 Biotinylated protein (BT202, 100 ng/mL) binds to HEK293 human embryonic kidney cell line transfected with recombinant human CD25/IL-2R alpha. (A) Binding is completely blocked by 2.5 µg/mL of Human Anti-Human CD25/IL-2R alpha Monoclonal Antibody (Catalog # MAB10203) but not by (B) Rabbit IgG Isotype Control (MAB1050) at the same concentration. Ligand binding was detected by staining cells with APC-conjugated Streptavidin (F0050).

Background: CD25/IL-2R alpha

Human IL-2 receptor alpha (IL-2R alpha ), also known as Tac antigen and as CD25, was initially identified as a 55 kDa membrane glycoprotein that is capable of binding IL-2. The IL-2R alpha cDNA encodes a 272 amino acid residue precursor Type I membrane protein with a 21 residue signal peptide, a 219 residue extracellular region, a 19 residue transmembrane region and a 13 residue cytoplasmic domain. IL-2R alpha lacks structural features characteristic of members of the cytokine receptor superfamily. By itself, IL-2R alpha binds IL-2 with low affinity. However, when IL-2R alpha is associated with the IL-2R beta and gamma chains, a high affinity heterotrimeric receptor complex that transduces IL 2 signals is formed.
