Background: DLL3
Delta-like 3 (DLL3) is a transmembrane Delta-like protein that contains one DSL domain and six EGF-like repeats. DLL3 is critical for somite segmentation and neurogenesis during early embryonic development. It does not bind Notch receptors but functions as an antagonist of Notch signaling. Defects in DLL3 cause axial skeletal abnormalities in spondylocostal dysostosis. Within the extracellular domain, human DLL3 shares 84% amino acid sequence identity with mouse and rat DLL3.

Detection of DLL3 on lung cancer cell line NCI-H82. NCI-H82 cells were treated with 100 nM of the human DLL3 antibody (catalog # MAB4315) or mouse IgG2b isotype control, followed by a secondary antibody goat a-mouse IgG Fc APC. Isotype control (RED), human DLL3 antibody (BLUE).
Recombinant DLL3 ECD Fc fusion was expressed transiently in 293. Generated conditioned media was run over Protein A column, then desalted, then polished on SEC column. The picture shown is a Western Blot for the collected SEC fractions, using MAB4315 as the primary antibody, followed by an anti-murine secondary antibody.
