Mitogen- and stress-activated protein kinase 1 (MSK1), also known as ribosomal protein S6 kinase 5 (RPS6KA5), belongs to the AGC family of kinases. Both MSK1 and the related MSK2, also known as RSKB and RPS6KA4, have two kinase domains connected by a regulatory linker region, and are activated by the mitogen-activated protein kinases ERK1, ERK2, and p38. Nuclear MSK phosphorylates and activates a number of transcription factors, including ATF1 and CREB.

MSK2/RSKB was detected in immersion fixed paraffin-embedded sections of human pancreas using Rabbit Anti-Human/Mouse Phospho-MSK2/RSKB (S196) Antigen Affinity-purified Polyclonal Antibody (Catalog # AF189) at 15 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Rabbit HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS005) and counter-stained with hematoxylin (blue). Detection of Human Phospho-MSK2/RSKB (S196) by Western Blot.
Western blot shows lysates of HepG2 human hepatocellular carcinoma cell line untreated (-) or treated (+) with 100 ng/mL Recombinant Human HGF (Catalog # 294-HG) for 15 minutes. PVDF membrane was probed with 0.5 µg/mL of Rabbit Anti-Human/Mouse Phospho-MSK2/RSKB (S196) Antigen Affinity-purified Polyclonal Antibody (Catalog # AF189), followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
