Detection of Human Ovastacin by Western Blot. Western blot shows lysates of K562 human chronic myelogenous leukemia cell line and OVCAR-3 human ovarian carcinoma cell line. PVDF membrane was probed with 1:1000 dilution of Rabbit Anti-Human Ovastacin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8549) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band was detected for Ovastacin at approximately 50 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Detection of Human Ovastacin by Simple WesternTM. Simple Western lane view shows lysates of K562 human chronic myelogenous leukemia cell line and OVCAR‑3 human ovarian carcinoma cell line, loaded at 0.2 mg/mL. Specific bands were detected for Ovastacin at approximately 69 kDa (as indicated) using 1:100 dilution of Rabbit Anti-Human Ovastacin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8549). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system. Non-specific interaction with the 230 kDa Simple Western standard may be seen with this antibody.

Background: Ovastacin
Ovastacin (ASTL, astacin-like metallo-endopeptidase and Oocyte astacin) is a 50 kDa member of the peptidase M12A family of proteins. Ovastacin is an oocyte-specific oolemmal receptor involved in sperm and egg adhesion and fertilization, it plays a role in the polyspermy inhibition. Human Ovastacin is a metalloendopeptidase that contains three zinc binding domains. Ovastacin cleaves the sperm-binding ZP2 at the surface of the zona pellucida after fertilization and cortical granule exocytosis, rendering the zona pellucida unable to support further sperm binding. Over aa 24-431, human Ovastacin shares 68% aa identity with mouse Ovastacin.
