Detection of Human PDZD2 by Western Blot. Western blot shows lysates of human prostate tissue. PVDF membrane was probed with 2 µg/mL of Mouse Anti-Human PDZD2 Monoclonal Antibody (Catalog # MAB7670) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for PDZD2 at approximately 37 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
PDZD2 in PC‑3 Human Cell Line. PDZD2 was detected in immersion fixed PC-3 human prostate cancer cell line using Mouse Anti-Human PDZD2 Monoclonal Antibody (Catalog # MAB7670) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red, upper panel; Catalog # NL007) and counterstained with DAPI (blue, lower panel). Background: PDZD2
PDZD2 (PSD95, Discs-large and ZO-1 domain-containing protein 2), also called AIPC (activated in prostate cancer) and PDZK3, is a multi-PDZ-domain protein that is ubiquitously expressed. The 2839 amino acid (aa) human PDZD2 is found in the endoplasmic reticulum. Caspase cleavage produces the 347 aa, 37 kDa secreted mature form (aa 2493-2839). A 2640 aa isoform is lacking 199 aa N-terminal to the mature sequence. The processed, mature form contains two PDZ domains which interact with the C-terminus of partner proteins, promotes maturation of pancreatic b cells, and is highly expressed in human prostate tumors. Human PDZD2 shares 72% aa sequence identity with mouse or rat PDZD2.
