In a functional ELISA, 1-5 μg/mL of this antibody will block 50% of the binding of 2 μg/mL Recombinant Human PILR‑ alpha (Catalog # 6484-PR) to immobilized recombinant human PANP Fc Chimera coated at 5 μg/mL (100 µL/well).

PILR-alpha binding to PANP blocked by PILR-alpha Antibody. In a functional ELISA, Recombinant Human PILR-a (Catalog # 6484-PR) binds to immobilized recombinant human PANP Fc Chimera coated at 5 µg/mL (100 µL/well) in a dose-dependent manner (orange line). Binding is blocked (green line) by increasing concentrations of Rabbit Anti-Human PILR-a Monoclonal Antibody (Catalog # MAB64842). At 1-5 µg/mL, this antibody will block 50% of the binding.

Detection of PILR- alpha R in HEK293 Human Cell Line Transfected with Human PILR-alpha and eGFP by Flow Cytometry. HEK293 human embryonic kidney cell line transfected with either (A) human PILR- a or (B) irrelevant transfectants and eGFP was stained with Rabbit Anti-Human PILR- a Monoclonal Antibody (Catalog # MAB64842) followed by Allophycocyanin-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # F0111). Quadrant markers were set based on control antibody staining (Catalog # MAB1050).

Background: PILR-alpha

PILR-alpha (paired immunoglobulin-like, type 2 receptor alpha) is a 44-50 kDa type I transmembrane (TM) paired receptor glycoprotein that belongs to the Ig Superfamily. It is expressed by monocytes, macrophages, CD14+CD1a- DC and retinal pigment cells, and is known to bind to CD99 and PANP. PILR-alpha acts as a receptor for HSV and serves as a negative immunomodulator that contains an ITIM. Mature human PILR-alpha is 284 amino acids (aa) in length. It contains one V-type Ig-like domain in its extracellular region (aa 32-150), and two ITIMs in its cytoplasmic domain (aa 267-272 and 296-301). There are multiple potential splice variants. One is TM and possesses a 35 aa substitution for aa 264-303, while others are soluble, and show a deletion of aa 152-224 that may be coupled to the 35 aa substitution noted above, or simply exhibit a 24 aa substitution for aa 152-303. Over aa 20-196, human PILR-alpha shares only 42% aa identity with mouse PILR-alpha, and 89% aa identity with human PILR-beta.
