Carcinoembryonic Antigen-Related Cell Adhesion Molecules (CEACAMs)

Carcinoembryonic Antigen-Related Cell Adhesion Molecules (CEACAMs) form part of the CEA protein family consisting of CEACAMs and the pregnancy-specific glycoproteins (PSGs). CEACAMs are Ig superfamily proteins encoded by 12 genes on human chromosome 19q13 and display diverse evolution, with only 5 CEACAMs conserved among mouse, rat, and human.

Originally described as a biomarker for colorectal cancer, CEACAMs have now been associated with a wide range of intracellular signaling processes and specific expression patterns. CEACAM-3 was identified as critical for opsonin-independent phagocytosis and bacterial clearance, whereas CEACAM-16 has been linked to autosomal dominant nonsyndromic deafness (ADNSHL). Additionally, CEACAM-1, CEACAM-5, CEACAM-6, and CEACAM-19 have been studied for their role in multiple cancers, with special interest as CAR-T targets. CEACAM family members have been shown to bind each other and have been described as receptors for Tim-3, galectins, and integrins.

CEACAM-21 Binds to Galectin-3. When Recombinant Human Galectin-3 (Catalog # 8259-GA) is coated at 1 ug/mL, Recombinant Human CEACAM-21 Fc Chimera (Catalog # 9870-CM) binds with an ED50 approximately 150 ng/mL. CEACAM-3 Binds to CEACAM-7. When Recombinant Human CEACAM-3/CD66d (Catalog # 9868-CM) is coated on to a tissue culture plate at 2 ug/mL Recombinant Human CEACAM-7 (Catalog # 9010-CM) binds with an ED50 of approximately 1 ug/mL. CEACAM-19 enhances adhesion of mouse fibroblasts. Immobilized Recombinant Human CEACAM-19 (Catalog # 9397-CM) increases cell adhesion of mouse fibroblast L cells. The ED50 for this effect is 0.4-2.4 μg/mL.
