Detection of Human Phospho-GSK‑3 beta (S9) by Western Blot. Western blot shows lysates of untreated (-) or MCF-7 human breast cancer cell line treated (+) with 100 ng/ml Recombinant Human IGF-I (Catalog # 291-G1) for 60 minutes and HeLa human cervical epithelial carcinoma cell line treated (+) with 200 nM PMA for 20 minutes. PVDF membrane was probed with 0.5 µg/mL of Rabbit Anti-Human Phospho-GSK-3 beta Monoclonal Antibody (Catalog # MAB8934) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band was detected for Phospho-GSK-3 beta (S9) at approximately 48 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Phospho-GSK-3 beta (S9) in MCF‑7 Human Cell Line. GSK-3 beta phosphorylated at S9 was detected in immersion fixed MCF-7 human breast cancer cell line untreated (lower panel) or treated with 100 ng/mL Recombinant Human IGF-I (Catalog # 291-G1; upper panel) using Rabbit Anti-Human Phospho-GSK-3 beta Monoclonal Antibody (Catalog # MAB8934) at a 1 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Rabbit IgG Secondary Antibody (red; Catalog # NL004) and counterstained with DAPI (blue). Detection of Human Phospho-GSK‑3 beta (S9) by Simple WesternTM. Simple Western lane view shows lysates of MCF-7 human breast cancer cell line untreated (-) or treated (+) with 100 ng/mL Recombinant Human IGF-I (Catalog # 291-G1) for 60 minutes, loaded at 0.2 mg/mL. A specific band was detected for Phospho-GSK-3 beta (S9) at approximately 56 kDa (as indicated) using 5 µg/mL of Rabbit Anti-Human Phospho-GSK-3 beta Monoclonal Antibody (Catalog # MAB8934). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system. Non-specific interaction with the 230 kDa Simple Western standard may be seen with this antibody.

Glycogen Synthase Kinase-3 (GSK-3) is a serine/threonine kinase initially identified asan inhibitor of glycogen synthase. Two isoforms (GSK-3 alpha and GSK-3 beta ) share 85% amino acid identity. GSK-3 beta, inhibited by phosphorylation at S9 by Akt, is involved in energy metabolism, body pattern formation, and neuronal cell development.
