APLP‑1 in SH‑SY5Y Human Cell Line. APLP‑1 was detected in immersion fixed SH‑SY5Y human neuroblastoma cell line (positive staining) and MOLT‑4 human acute lymphoblastic leukemia cell line (negative staining) using Mouse Anti-Human APLP‑1 Monoclonal Antibody (Catalog # MAB3129) at 25 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; NL007) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm. Staining was performed using our protocol for Fluorescent ICC Staining of Non-adherent Cells.

Background: APLP-1
APLP-1 is a transmembrane metalloprotein that is expressed in central neurons. Similar to APP and APLP-2, APLP-1 is susceptible to cleavage by various secretases, generating multiple fragments from the extracellular and intracellular domains. These include peptides similar to the amyloidogenic A beta peptides and a cytoplasmic fragment that associates with Fe65 family proteins and functions as a transcriptional activator. The extracellular domain contains heparin and collagen binding regions and shares 89% amino acid sequence identity between human and mouse.
