Hepassocin/FGL1 in HepG2 Human Cell Line. Hepassocin/FGL1 was detected in immersion fixed HepG2 human hepatocellular carcinoma cell line (positive stain) and absent in K562 human leukemia cell line (negative stain) using Mouse Anti-Human Hepassocin/FGL1 Monoclonal Antibody (Catalog # MAB16142) at 8 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; NL007) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm. Detection of Hepassocin/FGL1 in HepG2 Human Cell Line by Flow Cytometry. HepG2 human cell line was stained with Mouse Anti-Human Hepassocin/FGL1 Monoclonal Antibody (Catalog # MAB16142, filled histogram) or isotype control antibody (MAB002, open histogram), followed by Phycoerythrin-conjugated Anti-Mouse IgG Secondary Antibody (F0102B). To facilitate intracellular staining, cells were fixed and permeabilized with FlowX FoxP3/Transcription Factor Fixation & Perm Buffer Kit (FC012). Hepassocin, also known as hepatocyte-derived fibrinogen-related protein 1 (HFREP-1) and FGL1, is a liver-specific secreted protein belonging to the fibronogen superfamily, whose members share a fibrinogen domain at their C-termini. Hepassocin may play a role in the development of hepatocellular carcinomas.
