Detection of Human Nrf2 by Western Blot. Western blot shows lysates of HepG2 human hepatocellular carcinoma cell line untreated (-), treated (+) or mock treated (m) with 10 µM MG132 (Catalog # 1748) for 4 hours and 10 pmol siRNA. PVDF membrane was probed with 1 µg/mL of Mouse Anti-Human Nrf2 Monoclonal Antibody (Catalog # MAB3925) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for Nrf2 at approximately 110 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Detection of Human Nrf2 by Western Blot Confirmation of the IPA-predicted NRF2 activation in cancer cell lines by Western blot. Western blot analysis with NRF2-specific antibody in four different cell extracts per cell line (SiHa, HeLa, C33A, and HCK1T), corresponding to four biological replicates. Twenty μg was loaded. (a) A protein band of 70 kDa corresponding to NRF2 is detected. Immunoblotting for alpha -tubulin (50 kDa) was applied to ensure the comparable loading of proteins in each lane. Fold expression of NRF2 was assessed relative to HCK1T. (b) The mean NRF2 values for SiHa, HeLa, and C33A were 1.4 ± 0.1 (p < 0.05), 1.1 ± 0.2 (p > 0.05), and 2.3 ± 0.3 (p < 0.05) compared to HCK1T, respectively. Representative images of two biological replicates are shown for each cell line. Background: Nrf2

NF-E2 related factor 2 (Nrf2) is a member of the cap ‘n’ collar family of basic leucine zipper transcription factors. By binding antioxidant response elements (AREs), Nrf2 activates the transcription of genes encoding antioxidant and phase II drug-metabolizing enzymes needed to protect cells against oxidative damage. Transcriptional targets regulated by Nrf2 include genes encoding glutathione S-transferases, superoxide dismutase-1, and heme oxygenase-1.

RMC-1 cells, rat Muller cells, were cultured and treated, collect with NP-40 buffer, run WB. The problem is the molecular weight, we only see two specific bands around 75 KD marker, we didn't see any band around 120Kd as datasheet from RnD even longer exposure time. We would like to know why. I would like to provide more detailed information if you want to know. Thanks a lot.
