Detection of PMEL in Human Melanoma. PMEL was detected in immersion fixed paraffin-embedded sections of Human Melanoma using Mouse Anti-Human PMEL Monoclonal Antibody (Catalog # MAB11439) at 5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC001). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using VisUCyte Antigen Retrieval Reagent-Basic (Catalog # VCTS021). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm in cancer cells. Preparation and Storage

Background: PMEL
Premelanasome protein (PMEL) is a nonmutated "self" antigen expressed by melanocytes, most melanoma cells, substantia nigra, and retinal pigmented epithelium. This protein plays an essential role in the structural organization of premelanosomes. PMEL is over-expressed in all stages of melanoma progression and is negative prognostic marker for the overall survival rate of melanoma patients. PMEL is also associated with melanin-based pigmentation, neurodegenerative disease (functional amyloid) and glaucoma (missense mutations). How PMEL amyloid is tolerated within cells is of major therapeutic interest as it is related to Alzheimer's and Parkinson's disease.
