Detection of Human ZMIZ1/Zimp10 by Western Blot. Western blot shows lysates of LNCaP human prostate cancer cell line. PVDF membrane was probed with 1 µg/mL of Sheep Anti-Human ZMIZ1/Zimp10 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8107) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for ZMIZ1/Zimp10 at approximately 120-130 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
ZMIZ1/Zimp10 in Human Ovary. ZMIZ1/Zimp10 was detected in immersion fixed paraffin-embedded sections of human ovary using Sheep Anti-Human ZMIZ1/Zimp10 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8107) at 5 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Sheep HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS019) and counterstained with hematoxylin (blue). Specific staining was localized to nuclei. Background: ZMIZ1/Zimp10
Human ZMIZ1 (zinc finger and Miz-domain containing 1), previously called Zimp10 (zinc finger, Miz1, PIAS-like protein on chromosome 10) is a 1067 amino acid (aa), approximately 130 kDa cytoplasmic and nuclear protein. It is a coactivator of the androgen receptor (AR), and enhances AR-mediated transcription and AR sumoylation. It also coactivates other transcription factors such as SMAD4 and p53. It is normally expressed in human ovary, testis and prostate epithelial cells. The ZIMZ1 gene can be fused to ABL1 in a B-cell acute lymphocytic leukemia. Within the region used as an immunogen, human ZMIZ1 shares 98% and 97% aa sequence identity with mouse and rat ZMIZ1, respectively. A 997 aa isoform with an alternate sequence replacing aa 1-94 has been described.
