R&D Systems Proteins for Organoid Culture are Frequently More Active than Leading Competitor’s Proteins and Display High Lot-to-Lot Consistency
R&D Systems Recombinant Human Noggin is 30-fold More Active than Recombinant Human Noggin from a Leading Competitor and Displays Minimal Lot-to-Lot Variability. (A) Using the ATDC5 mouse chondrogenic cell line, the ability of R&D Systems Recombinant Human Noggin (Catalog # 6057-NG; orange line) or a top competitor’s recombinant human noggin (green line) to inhibit alkaline phosphatase production stimulated by Recombinant Human BMP-4 (R&D Systems, Catalog # 314-BP; 50 ng/mL) was assessed. The ED50 for R&D Systems Recombinant Human Noggin using this assay was approximately 30-fold higher than Noggin obtained from a leading competitor. (B) Using the same assay described in part (A), the lot-to-lot consistency of R&D Systems Recombinant Human Noggin (Catalog # 6057-NG) was assessed by side-by-side testing of the bioactivity of three independent lots of the protein (purple, light blue, orange trace on the graph). The dark blue trace on the graph is showing the ability of Recombinant Human BMP-4 (R&D Systems, Catalog # 314-BP; 50 ng/mL) to induce alkaline phosphatase production in the ATDC5 mouse chondrogenic cell line. (C, D) BG01V human embryonic stem cells were cultured in Mouse Embryonic Fibroblast Conditioned Media supplemented with FGF basic (5 ng/mL). Stem cells were driven into early cells of the neuroectoderm using a 3-day incubation in Recombinant Human Noggin (25 ug/mL) from either R&D Systems (Lot 1, Lot 2; Catalog # 6057-NG) or from two different competitors (Competitor 1, Competitor 2). Control cells were incubated in media without Noggin (No Noggin). The cells were stained for the early ectoderm marker, Otx2, and the neuroectoderm marker, SOX1. (C) Representative images of Otx2 (red), SOX1 (green), and DAPI (blue) staining in embryonic stem cells differentiated with Noggin from R&D Systems or Noggin from Competitor 2. (D) SOX1+ clusters were quantified under each of the indicated culture conditions. Cells treated with R&D Systems Noggin showed an increase in SOX1+ cells compared to both untreated and competitor-treated cells. R&D Systems Noggin showed consistent differentiation across the lots tested. BG01V human embryonic stem cells were licensed from ViaCyte, Inc.
Human Lung Organoids Embedded in Cultrex UltiMatrix RGF Basement Membrane Extract and Cultured Using R&D Systems Proteins. Adult stem cells isolated from human lung biopsy tissue were embedded in Cultrex UltiMatrix RGF Basement Membrane Extract (Catalog # BME001-05) and cultured for 20-60 days in lung organoid expansion medium, which includes Recombinant Human FGF-10 (Catalog # 345-FG) and Recombinant Human Noggin(Catalog # 6057-NG), along with the other reagents listed in the lung organoid expansion medium recipe in the human lung organoid culture protocol. Lung organoids were able to differentiate and exhibit markers for various cell types of the lung. Lung organoids were stained with (A) a Rabbit Anti-Human Cytokeratin 5 (KRT5) Monoclonal Antibody (Novus Biologicals, Catalog NB110-56916; green) and a Goat Anti-Human p63/TP73L Polyclonal Antibody (Catalog # AF1916; red) to visualize basal cells, (B) a Hamster Anti-Mouse Podoplanin (PDPN) Monoclonal Antibody (Novus Biologicals, Catalog # NB600-1015; green) to visualize alveolar type I cells and a Goat Anti-Human p63/TP73L Polyclonal Antibody (Catalog # AF1916; red) to visualize basal cells, and (C, D) a Mouse Anti-MUC5AC Monoclonal Antibody (Novus Biologicals, Catalog # NBP2-15196; green) to visualize Goblet cells and a Mouse Anti-Human/Mouse/Rat SOX2 Monoclonal Antibody (Catalog # MAB2018; red). All samples were counterstained with DAPI (Catalog # 5748; blue).
