Detection of Human THAP11 by Western Blot. Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line. PVDF Membrane was probed with 2 µg/mL of Mouse Anti-Human/Mouse THAP11 Monoclonal Antibody (Catalog # MAB5727) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for THAP11 at approximately 47 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
THAP11 in BG01V Human Stem Cells. THAP11 was detected in immersion fixed undifferentiated BG01V human embryonic stem cells using Mouse Anti-Human/Mouse THAP11 Monoclonal Antibody (Catalog # MAB5727) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (yellow; Catalog # NL007) and counterstained with DAPI (blue). Specific staining was localized to nuclei. Detection of Human THAP11 by Simple WesternTM. Simple Western lane view shows lysates of BG01V human embryonic stem cells, loaded at 0.2 mg/mL. A specific band was detected for THAP11 at approximately 56 kDa (as indicated) using 20 µg/mL of Mouse Anti-Human/Mouse THAP11 Monoclonal Antibody (Catalog # MAB5727). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

Background: THAP11
THAP11, also known as Ronin, is a 36-44 kDa member of the thanatos-associated protein family. It is ubiquitously expressed and through its repression of c-Myc transcription, blocks cell proliferation. Human THAP11 is 314 amino acids (aa) in length. It contains a DNA-binding THAP zinc-finger domain (aa 5-81), an NLS (aa 98-104), a polyGln region (aa 104-146), an Ala-rich segment (aa 197-221) and a C-terminal coiled-coil region (aa 255-305). THAP11 is phosphorylated on Thr25. Over aa 226-314, human and mouse THAP11 are 100% identical in amino acid sequence.
