CyTOF®, also known as mass cytometry, takes advantage of the low spectral overlap between heavy metal isotopes. In this technique, heavy metals are conjugated to antibodies, and their unique mass signatures are detected by time-of-flight mass spectrometry. Heavy metals take the place of fluorophores which are relatively limited by their overlap in fluorescence emission spectra. This allows the simultaneous detection and resolution of more molecular targets than are practical with the use of fluorescent flow cytometry dyes. Over 40 cellular targets can be identified on a single cell without the need for fluorescence compensation adjustments.
