Detection of FOLR1 in MCF‑7 Human Cell Line by Flow Cytometry. MCF-7 human breast cancer cell line was stained with Goat Anti-Human FOLR1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5646, filled histogram) or control antibody (AB-108-C, open histogram), followed by Allophycocyanin-conjugated Anti-Goat IgG Secondary Antibody (F0108).
Detection of Human FOLR1 by Western Blot. Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human FOLR1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5646) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (HAF017). A specific band was detected for FOLR1 at approximately 37 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Western Blot Shows Human FOLR1 Specificity by Using Knockout Cell Line. Western blot shows lysates of HeLa human cervical epithelial carcinoma parental cell line and FOLR1 knockout HeLa cell line (KO). PVDF membrane was probed with 1 µg/mL of Goat Anti-Human FOLR1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5646) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (HAF017). A specific band was detected for FOLR1 at approximately 40 kDa (as indicated) in the parental HeLa cell line, but is not detectable in knockout HeLa cell line. GAPDH (AF5718) is shown as a loading control. This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: FOLR1
Folate Receptor 1 (FOLR1), also known as Folate Receptor alpha and Folate Binding Protein (FBP), is a 37‑42 kDa protein that mediates the cellular uptake of folic acid and reduced folates. Dietary folates are required for many key metabolic processes including nucleotide and methionine synthesis, the interconversion of glycine and serine, and histidine breakdown (1, 2). Mature FOLR1 is an N-glycosylated protein that is anchored to the cell surface by a GPI linkage (3‑6). Human FOLR1 shares 83% amino acid sequence identity with mouse and rat FOLR1. FOLR1 is predominantly expressed on epithelial cells and is dramatically upregulated on many carcinomas (7, 8). It is critically required during early embryogenesis as shown in knockout mice which die in utero with gross morphological defects (9). FOLR1 is internalized to the endosomal system where it dissociates from its ligand before recycling to the cell surface (6, 10). A soluble form of FOLR1 can be proteolytically shed from the cell surface into the serum and breast milk (11).
